摘要:
短葶山麦冬为重要的园林观赏植物和药用植物,目前野生资源破坏严重,而通过组织培养技术可以获得大量种苗,满足市场需求。为解决短葶山麦冬组织培养存在消毒难的问题,以短葶山麦冬花蕾为外植体,通过对不同消毒方法、不同的培养基配方及不同移栽基质的筛选,建立了短葶山麦冬组培快繁体系。结果表明,75%酒精消毒30 s,无菌水清洗3遍,0.1%升汞溶液浸泡10 min,再用无菌水清洗5遍的消毒效果最好。最佳初代培养基是MS+1 mg·L-1 6-BA+0.2 mg·L-1 NAA,最佳继代培养基为MS+4 mg·L-1 6-BA+0.2 mg·L-1 NAA,最佳生根培养基为1/2 MS+1 mg·L-1 IBA+30 g·L-1蔗糖。红壤土、珍珠岩、沙的混合比为6∶3∶1时的成活率最高达到93.33%。
Abstract:
Liriope muscari (Decne.) Bailey is a kind of ornamental plants and medicinal plants, and its wild resource has been damaged severely.  So its seedlings have to be obtained by tissue culture to meet the market demand. In order to solve the problem of disinfection, a tissue culture system of L.muscari with its pedicles was built by screening different disinfection methods, different culture medium formulas and different matrixes for transplanting. Experimental results showed as follows. The most suitable procedure for sterilization was to soak the plants into 0.1% HgCl2 solution for 10 min and then wash them with sterile water for 5 times after soaking them into 75% alcohol disinfection for 30 s and washing with sterile water for 3 times. The optimum initial medium was MS+1 mg·L-16-BA + 0.2 mg·L-1NAA, the optimum subculture medium was MS+4 mg·L-16-BA + 0.2 mg·L-1NAA, and the optimum rooting medium was 1/2 MS+1 mg·L-1IBA +30 g·L-1 sucrose. The survival rate up to 93.33% when the ratio of red loam, perlite, sand mixing was 6∶3∶1.
