以多年生千层金植株为材料,采用小拱棚扦插育苗技术,通过L9(33)正交试验研究不同基质(草炭、蛭石、珍珠岩的不同质量配比)、生根剂浓度(100、200、400 mg·L-1)及处理时间(1、2、3 h)对千层金扦插生根率、平均根长、根系活力和叶绿素含量的影响。采用极差分析法和主成分分析法,初步筛选合适千层金扦插的基质配方、生根剂浓度和处理时间。结果表明:处理组合A1B2C3(基质为蛭石,在浓度为200 mg·L-1生根剂中处理3 h)扦插苗的生根率、根系活力和叶绿素含量均达到最高,分别为78.00%、63.69 mg·g-1·h-1、13.16 mg·g-1;处理组合A2B1C3(基质为m草炭m蛭石=1:1,在浓度为100 mg·L-1生根剂中处理3 h)扦插苗的平均根长最长,为10.00 cm。主成分分析表明,生根率是影响千层金扦插效果的主要指标,基质是影响千层金扦插成活及生根率的最关键因素。处理组合A1B2C3的综合得分最高,为1.06,是千层金扦插效果最好的组合。
Abstract:
Perennial Melaleuca bracteata branch cuttings are used as cutting seedlings of small arch shed by adopting cutting seedling technology. An orthogonal experimental design was used to study the effects of substrates(different ratios of peat, vermiculite, and perlite) and growth regulator concentrations(100, 200, 400 mg·L-1) on the survival rate and processing time, the rooting rate, chlorophyll content, and root vitality of the cuttings. The effects of the concentration ratio of rooting agents and processing time on rooting of M. bracteata cuttings were studied using an orthogonal experimental design and principal component analysis. The results showed that the rooting rate, root vitality, and chlorophyll content of cutting seedlings reached the highest levels of 78.00%, 63.69 mg·g-1·h-1, 13.16 mg·g-1, respectively, with the treatment combination A1B2C3 where the substrate was vermiculite, the cuttings were treated with a rooting agent concentration of 200 mg·L-1 for 3 h. The average root length of cutting seedlings reached the longest, at 10.0 cm, with the treatment combination A2B1C3(the cuttings were treated with 100 mg·L-1 rooting agent for 3 hours, and then cut into a mixed medium of peat soil: vermiculite at a ratio of 1:1). Principal component analysis showed that the rooting rate is the main indicator of the effect of M. bracteata cuttings, and the substrate is the most critical factor affecting the survival and rooting rate. The treatment group treated with vermiculite as the substrate and soaked with 200 mg·L-1 rooting agent concentration for 3 hours had the highest comprehensive score, which was 1.06. Therefore, A1B2C3 is the optimal treatment combination.
